PRINCIPLE OF THE TEST.
Fixation, decalcification, and staining of bone marrow.
SPECIMEN REQUIRED.
All human tissue excised at surgery, outpatient clinics,
and postmortems, fresh or in fixative, along with a filled-out
Tissue Examination Form
(U. S. Standard Form 515, USSF515).
REAGENTS, INSTRUMENTATION.
Fixative.
70% alcohol,
Decalcifying solution (RDO),
Flex.
STEP-BY-STEP DESCRIPTION.
Specimen will be logged in and dictated
by the resident on the day that it
arrives in the histology laboratory.
BIOPSY:
1. Allow to remain in fixative overnight.
2. In the morning, pour off fixative and rinse in 70% ethanol several times.
3. Pour on decalcifying solution. Allow to remain for one hour.
4. Pour off decal. Rinse with 70% ethyl alcohol.
5. Place on blue sponge in yellow
cassette that is properly labeled.
6. Wash the remainder of the day in two changes of 70% ethanol
(in the beaker with the stir bar).
7. Put on with the surgicals at the end of the day.
8. Include a request for 10 LABELS WRIGHT STAIN
on the list of blocks submitted to histology.
These labels will subsequently be attached to the
aspiration slides when the bone marrow case is signed out.
ASPIRATE:
1. Gently agitate, in order to loosen all material.
2. Pour into a teabag. Rinse out
the container with 70% ethyl alcohol,
and pour that into the teabag. If necessary,
use an applicator stick to scrape
remaining material out of container
and into teabag.
3. Fold the teabag and place in yellow cassette
that has been properly labeled.
4. Wash for the rest of the day in 70% ethanol
(2 changes), and put on with the surgicals.
5. When aspirate smear is obtained, fix in 95% Flex, and let stand
until iron controls go through clearing into water. Then put iron control
and smear into the iron solution, and continue with the iron stain.
