PRINCIPLE OF THE TEST.

      To stain paraffinized tissue sections for mucin, required in the diagnosis of certain diseases encountered in anatomic pathology, especially poorly-differentiated adenocarcinomas.



SPECIMEN REQUIRED.

      All human tissue excised at surgery, outpatient clinics, and postmortems, fresh or in fixative, along with a filled-out Tissue Examination Form (U. S. Standard Form 515, USSF515).



REAGENTS, INSTRUMENTATION.


      Weigert's Iron Hematoxylin. Metanil Yellow Solution. Mucicarmine Stain. Absolute ethanol. 95% ethanol. Xylene or xylene-substitute.



STEP-BY-STEP DESCRIPTION.


FIXATION: 10% Buffered Formalin.

TECHNIQUE: Paraffin. Cut at 6 microns.

SOLUTIONS:

1. Weigert's Iron Hematoxylin

2. Metanil Yellow Solution.

3. Mucicarmine Stain.

STAINING PROCEDURE:

1. Deparaffinize sections, and hydrate to water.

2. Control slide must be used.

3. Place slides in Weigert's hematoxylin for 7 minutes.

4. Wash well in running water. At least 5 minutes.

5. Place slides in Metanil yellow for 1 minute.

6. Wash well in running water.

7. Mucicarmine stain (diluted 1 to 4) for 45 minutes.

8. When satisfactory, wash ftuickly in water.

9. Alcohol - 95% -- 2 changes.

10. Alcohol - 100% -- 2 changes.

11. Xylene -- 2 changes.

12. Mount with poly Mount or any other acceptable mounting medium.

RESULTS:

Mucin ............... Bright Rose

Nuclei .............. Black

Connective Tissue ... Yellow

THE ABOVE STAINS CAN BE PURCHASED FROM POLY SCIENTIFIC, READY FOR USE.
